A Study on the Cytotoxicity of Mitomycin C in Cultured Rat Fibroblasts

Abstract
Present study was carried out to investigate the cytotoxicity of mitomycin C in cultured rat fibroblasts. The colorimetric assays of neutral red(NR) and tetrazolium MTT and also the measurement of the rate of DNA synthesis and sister chromatid
exchange
(SCE) analysis were performed in cells cultured in the media containing various concentrations of mitomycin C.
@ES The results are as follows:
@EN 1. The uptake ability of NR was decreased dose-dependently and NR90 and NR50 values of mitomycin C were 0.5§¶/§¢and 2.3§¶/§¢ respectively.
2. Mitomycin C induced the dose-dependent decrease of the activity of mitochondrial succinic dehydrogenase and MTT90 and MTt50 values were 0.7§¶/§¢, respectively.
3. Mitomycin C inhibited DNA synthesis dose-dependently and the rates of DNA synthesis at concentrations of mitomycin C, 0.5§¶/§¢(NR90) and 10-3§¶/§¢were 2% and 50% of the control.
4. Frequency of sister chromatid exchange increased dose-dependently.
From above results, it suggests that mitomycin C induced the DNA damage at below concentrations which dose not cause cytotoxicity as determined by NR and MTT assays.
